Abstract
The objective of this study was to comparative various levels of benzylaminopurine (BAP) on the induction, the quantity and the length of shoot apices of Kluay Lep Mu Nang banana [Musa (AA group) Kluay Lep Mu Nang].
Seven different MS mediums with and without plant growth regulators were used to culture the youngest shoot apices. Basal MS medium was used as a control, whilst the rests of 6 MS medium treatments were supplemented with the equal amount of 0.2 mg/l Naphthalene Acetic Acid (NAA) and incremental levels of BAP as follows: 0.5, 1.0, 2.0, 3.0, 4.0 and 5.0 mg/l, respectively. Small pieces of sterile shoot apices of Kluay Lep Mu Nang were randomly cultured into a glass bottle containing medium treatment, which of 20 glasses bottle were used per treatment. The following 3 culturing periods consisted of the first period for shoot induction (30 days per period). The second and third period were continuously allocated using the same medium treatment to determine its shoot induction, shoot quantity and shoot length, respectively. The results in the first period showed that the induction of shoot obtained 1-3 shoots were not significantly differences among the medium treatments (p<0.05). This result implied that plant growth regulators were not affected at the early stage on the growing of shoot. However, the contradicted results were observed in the following period. The quantity of shoots in the second and the third period for the inclusion of 5.0 mg/l BAP treatment were 11 and 8 shoots per bottle. These values were respectively higher than those for all the rest of medium treatment. In contrast, the BAP least than 5 mg/l (0-4 mg/l) showed the enhancement of the shoot length for the second and the third period. Among the treatments, although the mean values of quantity and the length of shoots with the addition of 3.0 mg/l BAP were intermediate. However, based on our observation it can be recommended that the above BAP level was appropriate in sub-culturing for the plant micro-propagation in order to obtain the shoot healthy, shoot number as well as shoot length.
บทคัดย่อ
วัตถุประสงค์ของการศึกษาในครั้งนี้เพื่อเปรียบเทียบระดับการใช้ BAP ที่แตกต่างกัน ต่อการชักนำยอด จำนวนยอดและความยาวของยอดกล้วยเล็บมือนาง โดยใช้ชิ้นส่วนปลายยอดของหน่อกล้วยเล็บมือนางวางเลี้ยงบนอาหารสูตร MS ที่มีการเติมสารควบคุมการเติบโต NAA (Napthalene Acetic Acid) ความเข้มข้น 0.2 mg/l ร่วมกับ Benzylaminopurine (BAP) ในระดับความเข้มข้น 0.5, 1, 2, 3, 4, 5 mg/l โดยสูตรอาหาร MS ที่ไม่มีการเติมสารควบคุมการเติบโตเป็นชุดควบคุม นำปลายยอดหน่อกล้วยที่ผ่านการฆ่าเชื้อวางเลี้ยงในอาหารสูตรต่างๆ ทำการวางเลี้ยงและสุ่มเก็บผลจำนวน 20 ขวดต่อชุดการทดลอง การวางเลี้ยงแบ่งเป็น 3 ช่วงเวลา คือ วางเลี้ยงเพื่อชักนำยอด และการย้ายเลี้ยงเพื่อการขยายพันธุ์อีก 2 ช่วงเวลา ช่วงเวลาละ 30 วัน พบว่าในช่วงการชักนำยอดการเติมและไม่เติมสารควบคุมการเติบโตจะให้จำนวนยอดไม่แตกต่างกัน (p<0.05) ซึ่งทุกสูตรอาหารให้จำนวนยอดประมาณ 1-3 ยอด ทำการย้ายเลี้ยงยอดบนอาหารใหม่สูตรเดิมทุกเดือนเป็นระยะเวลา 2 เดือน พบว่า การย้ายเลี้ยงบนอาหารสูตร BAP 5 mg/l ได้จำนวนยอดมากที่สุด คือ 11 และ 8 ยอด เมื่อมีการย้ายเลี้ยงในเดือนที่ 1 และเดือนที่ 2 ตามลำดับ ในขณะที่การเติม BAPในปริมาณน้อยกว่า 5 mg/l (0.5-4 mg/l) มีแนวโน้มกระตุ้นในด้านความยาวของยอดมากกว่าจำนวนยอด เมื่อพิจารณาจำนวนยอดร่วมกับความยาวยอด พบว่า การเติม BAP 3 mg/l มีผลทำให้จำนวนยอดและความยาวยอดอยู่ในระดับที่เหมาะสม ดังนั้นสูตรอาหารที่เหมาะสมต่อการขยายพันธุ์กล้วยเล็บมือนาง คือ สูตรอาหารที่เติม BAP 3 mg/l ร่วมกับ NAA 2.0 mg/l ซึ่งมีผลต่อความสมบูรณ์ของยอด จำนวนยอด และความยาวของยอดต้นกล้วยเล็บมือนาง
